  • Simple Description: The goal of this project is to obtain additional preclinical data on the therapeutic efficacy and neurotoxicity of short term intracerebral (i.c.) convection enhanced delivery (CED) or prolonged infusion of carboplatin in combination with megavoltage X-irradiation for the treatment of the F98 rat glioma. These data will be directly relevant and supportive of a Phase I clinical trial (pre-IND #:102427), a synopsis of which is included as Specific Aim #2. The purpose of the clinical trial will be to establish the maximum tolerated dose and define the toxicity profile of carboplatin administered i.c. via CED in patients with glioblastoma multiforme (GBM) who are candidates for a second craniotomy following current standard therapy. Once the safety of i.c. CED of carboplatin has been demonstrated, then a second clinical trial will be initiated to evaluate the combination of CED of carboplatin and photon irradiation for the treatment of patients with recurrent glioblastomas (GBM).
